Thanks to all who replied. I followed everyone's suggestions with NO SUCCESS.
As instructed I did the following.
1. Uninstalled Microsoft AntiSpyware
2. Rebooted
3. Run Windows Uninstall Cleanup - which had no reference to Microsoft
AntiSpyware nor Windows Defender nor Defender Signatures
4. did the Msiexec /x {A5CC2A09-E9D3-49EC-923D-03874BBD4C2C} which came back
to say "This action is only valid for products which are currently installed."
5. I downloaded a new Windows Defender dated 2/23/2006 and installed it.
6. I clicked on check for updates and run quick install.

Same error. Windows Defender was unable to complete the update: 0x80070643
BTW. The Windows Defender screen said "signature version: 1.0.0.0"

MICROSOFT -- Please withdraw this software until it really works!

Yes, I agree with you... I posted this type of message previously and it
worked multiple times. I believe they were trying to fix the problem so we
wouldn't need to do this anymore. Perhaps they changed the hash on us...It
will come out soon. Don't you worry. In the mean time, until they come up
with the answer, what version were you updating from? Have you tried this way
before that it worked?

I have been using Microsoft AntiSpyware and it updates fine all by itself.

The installation for Windows Defender never worked for me.
Someone suggested that my problem was because I have my My Documents and my
Outlook files on a mapped drive.

I finally had success !
The problem was quite obscure.

The Application Event Log had the following entry:
Source: MsiInstaller, Event ID: 11327, User: NT Authority\SYSTEM, Error
Product: Windows Defender Signatures -- Error 1327.Invalid Drive: y:\

The Y: drive holds "My Documents" and my Outlook.pst files. This is a mapped
drive on our server which has automatic backups.

So I figured, let me try to install it from an administrator account which
does not have any mapped drives. Unfortunately same error! So now I'm really
mad, and decide to go nuclear.

I pull out my trusty Registry Cleaner and find some references to the Y:
drive even for this other user. Kaboom! Blasted those entries clear to Mars.

Now it installs like a kitten and updates and we're all good to go.
