Windows defender was unable to complete the update: 0x80240022.

when I tried to update my windows defender defnition it gives the message
"Windows defender was unable to complete the update: 0x80240022. The wrong
diskette is in the drive. Insert %2 (Volume Serial Number: %3) into drive
%1.".
I removed windows defender .executed the command
Msiexec /x {A5CC2A09-E9D3-49EC-923D-03874BBD4C2C}
deleted the folder C:\Documents and Settings\All
Users\Application Data\Microsoft\Windows Defender
and downloaded the latest version of defenderand installed .still I am
getting the same error message .
Now almost all the computer which I installed windows defender gives me same
error message.

The message Engel refers to is indeed about the message you've received.
However, the basic content is that Microsoft is aware of the problem and is
investigating.

Have you tried updating via Windows Update since you did the remove and
reinstall?

yes
if client configured for WSUS update it will give above error .when I
disable this group policy for WSUS no problem.

I've just posted the step by step for configuring WSUS to distribute Windows
Defender definitions several times today--check posts in this group, today,
with my name on them to get that information.

Windows Defender will get definitions from WSUS when WSUS is present on the
network, and configured to distribute those definitions. If it is not
configured to do so, the implication is that the application is not
approved, and Windows Defender will not update in that circumstance.

Disable Firewall!

Mine was the same problem. After de-install / re-install, finally came to the conclusion that the Kerio Personal Firewall might be blocking the update. Disabled it and the update went right through!

The configuration of the computer was such that it originally had the Microsoft Giant Antispyware on it. That seems to make a difference in this problem, but perhaps it is mostly a firewall one, as the settings are not obvious as to which applications are the ones for the Microsoft product.
