I don't know of a key command to open Magnifier in one go, but pressing
Windows Logo key + U opens the accessiblity Utilty Manager, from which you
can quickly start and stop magnifier (as well as the other accessiblity
options).

If you have a better solution let us know, I use magnifier myself and would
be interested.
