Can't get line in to work What port?

I have tried plugging an audio lead into both the line in of the
motherboard & into line in of my TV tuner card which also has analogue
in but I my PC can't hear anything at all. It's been suggested that I
don't have the correct port enabled. If this is so how do I determine
what port should be used & how do I enable it.

I do know that my audio is working because I get sound both from other
programs as well as the TV tuner card.

If you think the problem comes from some other cause please advise.

A line in connector is usually connect to a line out from the source.
