Build on One Motherboard -- Deploy to Another

I need to build a new system on a SATA drive for a machine with an AUSU A8N32-SLI DELUXE
motherboard. Unfortunately, I cannot take the machine down for the time it would take to do the
build.

I have a machine with an ASUS M3A78-EM motherboard. If I do the build on that machine and then run
sysprep will I be able to use the drive in the A8N32-DELUXE machine?

Probably not since the hw is different, eg sata controler
Are you not able to clone to your new drive on the existing sys?

Unless both boards have very similar SATA controllers it won't work, if they
do you may pull it off. The surefire way to have it work is to connect the
hard drive to a PCI SATA controller for the install, and move that
controller to the second machine, then switch the hard drive to the
motherboard controller after you've got all the drivers installed, I've had
excellent success with this method.
