If you're talking about selecting the Windows Update shortcut in the start
menu to access
http://susserver instead of
http://windowsupdate.microsoft.com then that wont work. SUS only works with
the automatic update client (see Laura's helpful posts on this). You cannot
have your SUS server run like an internal Windows Update Server through a
web browser - only via the clients. Whether or not you could write your own
web front-end for this is another matter...

By configuring Computer Configuration\Administrative Templates\Windows
Components\Windows Update in your Group Policy, what you're effectively
saying is:
"Hey! All you clients that this GPO applies to! When you need to go get
updates, use THIS server instead of going to Windows Update."
As long as your clients are SUS-aware, that's all it takes.
