Yes, you can do this. Some programs and utilties place files there while
they are active. If a program happens to need one or more of those files, it
will not let you delete that particular file. Th best time to do this is
right after a fresh reboot, before you open any programs.

You can delete anything in the temporary directorys and in some cases, any
file ending with .tmp.
Temporary directorys can be found in:
C:\WINDOWS\Temp
C:\Documents and Settings\Administrator\Local Settings\Temp
C:\Documents and Settings\Administrator\Local Settings\Temporary Internet
Files
Replace Windows with your windows directory or replace it with %windir% in
scripts.
Replace Administrator with the username or replace it with %username% in
scripts.
