Sites and Subnets

If you don't define any sites and subnets for your clients, but all the
Domain Controllers have been defined within a particular site/subnet, then
is there any negative impact to the client machines?

What if a client belongs to a site that has a DC assigned that same site,
but that DC is from a different domain than the client ?

What do you mean by a "site"? An address range? Or something else? These
are rather vague questions, perhaps you will get better help by trying to be
specific about what you're investigating and/or what problems you're having.

Sorry Richard, I was referring to Active Directory Sites, and the
configuration of those sites within the active directory environment.

No, as long as there aren't slow WAN links between the clients and the DCs.

If the DC isn't in the same Domain then it is irrelevant.
