Repeated Backup of PC - Vista Ultimate

I have already made a backup of my PC (and seprately set up a schedule for
file backups of data).
Because I have made some changes on my PC I now want to back that up.
If I just run the PC backup tool again will it create a separate, new 100%
backup of my system drive or will this be a differential backup?
Altenatively should I (Can I) delete the existing backup and start again?
What's the best approach please?
I didn't find what I needed on the "Help and How to"

If you just run the backup tool and don't change any settings then you get a
differential backup. You create a new 'backup set' by checking the 'Create a
new, full backup now ....' checkbox optioni via Backup settings (Control
Panel)

You can also delete the existing backup, by navigating to the drive and
opening the folder with the same name as your computer and deleting the
folder. Each 'backup set' is in a new folder there.
