Partition on Hard Drive

I have a single hard drive partitioned into drives C and D. I need to format
the C drive. Will I loose every thing on the D drive, if I leave the
partition intact?

No but...
If C has Windows installed on it and there are any applications installed on
D then they will become non-functional when you re-install Windows.
If however D only contains data files, photos, downloads, Etc. you will be
OK as long as you don't attempt to change partition sizes prior to
formatting the C partition.

In addition to what the other posters said, things can go wrong. Make sure
you have a full and complete backup of all important data. You should
always, always have a full and complete backup. Also make sure you have the
original media for any programs for installation. Sometimes things don't go
right.
