This is a symptom of a malware/virus infection, like Klez, Yaha, spybot,
gaobot, etc. You need to thoroughly scan your system, but some of these
will also disable antivirus apps.

Try restarting in Safe Mode and see if you can get into msconfig there.
You should be able to see and disable the launchers, but they are likely to
be recreated once you boot in regular mode. So, use the information you are
given to identify the particular worm and get the removal tool.

Also, get ccleaner, www.ccleaner.com and use it to clear out the temp
folders and browser caches. This may well help you get control of the
system.

Once you have some control and can connect to the internet, use Trend
Micro's Housecall to scan the system. housecall.trendmicro.com

The type of behavior you describe is typical behavior of more than
one virus/worm, the three below being the most common:

W32.Klez

W32.Yaha
http://securityresponse.symantec.com/avcenter/venc/data/[email protected]

W32.Spybot.Worm
http://securityresponse.symantec.com/avcenter/venc/data/w32.spybot.worm.html

Because many of the newer viruses and worms, such as the Spybot
mentioned above, can disable antivirus applications whose definitions
aren't kept up-to-date, try using one or more of the free on-line
scanners to double-check your system.

I've tried almost all suggested programs. Managed to find some
Malwares and Graywares but my taskmanager is still disabled and
situation with MSCONFIG and REGEDIT is the same. System is running
slow as well.

I have important data in my system so i can not format the disk,

The first thing to do would be to enable the registry editor, then you
should be able to work on the other problems.

Use a VBS script:

http://search.yahoo.com/search?ei=UTF-8&fr=yfp-t-501&cop=mss&p=enable+registry+tools+.vbs&x=0&y=0

The first hits point to Doug and Kelly's vbs solutions, both very
trustworthy sites.

This is another good site, they have a VBS script available as well as a
modified registry editor that works even when the restrictions are in
place:
