Why not just convert the drive to NTFS?

How to convert a FAT16 volume or a FAT32 volume to an NTFS file system in
Windows XP - http://support.microsoft.com/default.aspx?kbid=307881

If you cannot or will not do that, you may find some solutions that have
been produced by third-party companies. A search with a good
search-engine
should produce some relevant results with keywords 'secure folders fat32'

In order to "secure" the C drive, you will need to convert it to a file
system that has built in security features (NTFS). Backup your important
files/folder/emails/documents/pictures/etc onto your other drives (just in
case) and look into the command line "convert" command.
