Can I read files on my other computer that windows ME
while my main computer has NFTS drive configuations?

The os "reads" the file system.
If your booted into win xp it can see ntfs, fat, fat16, and fat32.
If your booted into win me it can see fat, fat16, fat32 drives.

If you're talking about over a network, the file system type has no bearing
whatsoever.

Yes, in both directions. The ME computer will be able to read anything
the XP machine shares (regardless of file system), and the XP machine
will be able to read anything the ME computer shares.

ME will make a request for a file over the network. The XP machine
will receive that request, read the file and send it. The ME machine
will receive the file from the network. It will not know or care what
file system the other machine uses.
