Microsoft spyware won't start

When I click the icon for the program to run it starts saying,"loading
spyware definitions" then it closes out.
I tried updating it but when i try to install the up date it pops up then
closes out also.

Dave's referenced an excellent scanner, but Microsoft also has an online
scanner which has their most up to date signatures:

click Protect in the left column, and then do a "protection scan."
