Can you protect a local drive so that users logging in to the machine
cannot access it unless they belong to a group that has rights to it?

TIA.

The drive(s) need to be NTFS format. You can set rights (privileges) to
folders, files even whole drives if you'd like.

if you are using an nt based os you can lock it down through ntfs
permissioning.

If someone is logged into the local machine you can still base your ntfs
permissions off of local users or groups. If it is really something you
want only yourself to be able to view then encrypt the files/folders with
EFS....go to the properties of a folder/file, then go to advanced and
encrypt. Then only that user can open it, or a recovery agent, but just
dont give the other users admin priveleges.
