I have created a new OU and move some of my users to the
new OU.
I have created a new GPO for the new OU.

The new GPO does not take effect unless I disable the
default domain GPO or move the computer account to the new
OU. I know that this is necessary if the changes are made
in the computer level but they were made at the user level.
I have also tried to put a tick in the block inheritance
but that does not help. also tried no override.

It sounds like you may have loopback processing enabled in the domain GPO. Look under
computer configuration/administrative templates/system/group policy to see if it is
enabled there. --- Steve

Thank you. That was it, I have never use loopback before,
now I understand it very well.
