How to get best results from Nikon Coolscan 5000?

I'm having trouble getting good quality scans from my Nikon Coolscan
5000.

I've tried to use both NikonScan and Vuescan, and have found that
NikonScan produces the sharpest scans, but the colour reproduction is
not particularly good. Vuescan on the other hand seems to get very
accurate colours, but the scans don't appear to be as sharp when
viewed side by side with the NikonScan output.

Has anyone else found this, and if you are getting good scans from
either package, what settings do you use?

Try moving the VueScan focus position (the blinking box in the preview).

Alternatively, try turning on the "Filter|Sharpen" option.
