how to count the total number of employee?

Maybe not, another equally viable method would be to stand someone at the
works entrance and count them as they arrive for work. If that person is an
employee they must; obviously, remember to count themselves and of course
make an allowance for anyone on sick/holiday leave and those working from
home.....

No, on reflection I agree a formula is probably best
