No. The question in your message body is not the same as the question
in your subject line.
1. You cannot cut/paste installed programs from the 40GB drive to the
80GB drive and have them work. You must uninstall/reinstall them.
Some files will always go to the system drive, however.
2. To use the 80GB drive as backup, you can either manually copy data
files there or use an automatic copy program such as Second Copy or
use an imaging/backup program such as Acronis True Image. "Backup"
implies redundancy in order to be able to restore the data.

There was a program called MagicMover that came with older versions of
Partition Magic that would do just that. It was very slick and worked
very well with Windows 9x systems. I never tried it on Windows XP
though, so I don't know if it will work there or not.
This is limited in what it can move, but this might help you out too.
