Yes with some limits.
You can move all of your documents over to the external hard drive.
You can uninstall programs, and then reinstall them on the external hard drive, but then don't
unplug it.
(you programs will run slower then)
You can clean up temporary files to give you some room.
You can specify the virtual memory to the external drive, again, don't unplug it.

But you will find that a much better solution is to buy a new internal hard drive, clone the old to
the new and swap them out.
All programs will install some parts on the C drive, even if you put the bulk of them on a non-C
drive.

Addendum:

If you don't (or rarely) use the hiberation feature, you can disable that as
well. This frees up disk space equal to the amount of RAM installed in your
computer.
