A friend of mine believes that all the files you have on your desktop e.g.
videos, are stored in RAM and affect performance. Surely its just a a pointer
stored in RAM to the file on the hard drive and not the whole file stored
there indefinately. Can anyone clarify?

Your friend is incorrect.
Prove it to them.
Copy a 2+GB file onto the desktop.
It's not using RAM.
