Customise auto replies in Outlook 2007

Is it possible to send an automated email reply to selective incoming emails?
Out of Office replies go to all incoming. I want to give a specific message
(not necessarily an Out of Office reply) to a few choice people.

Secondary to that question: can I prevent new emails from opening when I
move my cursor over it (using the up and down arrows on my keyboard) or when
I move it to my deleted items folder?

1) You can use a rule for that.

2) Note that the envelope is nothing more than an indication for your
convenience. It doesn't say anything about the mail being opened at a
technical level. That said, you can change the behavior of the unread/read
envelope in Tools-> Options-> tab Other-> button Reading Pane
