could not delete downloaded file from folder

downloaded an mpeg video file from a popular meta video site. Could not
delete file from system. Message appears:

cannot delete <filename> from: it is being used by another person or
program. Close any programs that might be using the file and try again.

Could it be a new trojan or spyware running in the background. I have done a
windows defender scan on hard drive and Norton anti-virus scan, negative
results.

While what you suggest is possible, it is also possible that you
are just seeing the effect of the delayed handle recycling in XP.
If the problem exists after a while, or after a reboot (some apps
are ill-behaved, like the Adobe pdf viewer for IE keeps its process
running after you have stopped using it, and you may have some
residual but innocent process still running without any window),
then you likely have an issue to investigate.
