I have
P4P800 Asus motherboard
2 drives X 2 partitions

If I add a serial ata drive what drive letter the SATA will get?

It can have anyone you want it to have (as long as it is not used by another
drive).
Just assign it a drive letter in the Disk Management section of the Computer
Management admin tool.

It shouldn't be "C:", if that is what you are worrying about.
Actually, it depends on:
1) Whether you have a Zip drive installed and connected when you
install the Serial ATA drive -- If such is the case, there is a good
chance that the new drive will take over the C: drive letter, so make
sure no Zip drives are physically connected when you install the
Serial ATA drive.
2) How many CD/DVD drives you have installed

For all intents and purposes, a Serial ATA drive will appear as a
normal IDE drive, and will be added to Windows as a normal IDE drive
would be added. This is the way mine appears on my P4P800 Asus
motherboard. I have a single hard drive, so my SATA drive appears as
a normal IDE drive, with the drive designation "C:"

If you are dissatisfied with the drive designation Windows gives the
SATA drive, you can change it yourself from within Windows Disk
Management.
