application won't run... 16 bit subsystem error msg

All of a sudden, a W2K workstation is posting the error below when launching
a program from the military called EPSQ. Security clearance app available
via www.dss.mil . Can't trace it back to anything installed since the failure
began. Anybody have a clue? Tx

16 bit Windows Subsystem
C:\WINNT\SYSTEM32\AUTOEXEC.NT. The system file is not suitable for running
MS-DOS and Microsoft Windows applications.
