After sending a message and clicking the "read receipt" box, how do you tell

if the message has been read?

Depends upon who you sent it to. Generally speaking if you send a read
receipt request and the other person "reads" the message using a client
that supports read receipts, and a server that supports read receipts, and
in an organization that doesn't block receipts to the Internet...then you
will get a message back acknowleging that the message has been "read."

However, there are a lot of situations where read receipts, particularly
to users out on the Internet, don't actually work. So I wouldn't depend
upon them.

And to add to what Ben said, there are those, like me, who turn off the
option wherever possible.
