You can but there isn't many places that you will see it (tool tips and a column in Detail View). There are also two ways of folder comments and XP only shows one way.

[.ShellClassInfo]
InfoTip=An optional description for the folder that displays in a pop up tip and web view when selected

Add this to the desktop.ini. Customise the folder first which will create the desktop.ini then edit it.

This won't be useful to me unless I can see the column in Detail View. How
am I able to see that, or is there another way of inserting a comment?

You didn't understand the simple question asked if you are sticking to your
original answer which makes your last comment kind of ironic... Don't tell
me I've got to start a 3rd thread because two people (MVP's!!!) give cryptic
answers and refuse to give a straight answer?

I added every available column in detail view (even all the stuff in
More...) and the infotip isn't there. The infotip works as a mouse hover.
I can add a summary comment to a normal file and it shows in detail view.
So what is it that you're saying I am not reading? There is no column in
detail view which displays infotip. You said a column in Detail View shows
infotip.

Is anyone else able to replicate David's instructions to make them work in
Windows XP?
