I had a Lexmark E321 that had been printing but now there
doesn't seem to be any connection between the computer
and the printer. I have tried everything from replacing
cable, to printing from command prompt, reinstalling
printers, drivers, etc. Everything appears to be working
in the device manager.

It's a Lexmark.

Follow Bruce Sanderson's instructions to COMPLETELY uninstall the printer
(having first disconnected it from the PC)

Then download a new set of drivers from www.lexmark.com and install them.
Do NOT connect the printer back to the PC until the software tells you to do
so.
