I have added a new route between my checkpoint firewall
which resides on a w2k server that route times out
although I have two other routes that working just fine.
Do I need to bounce anything to get this working?
 
No, you don't.

You don't give much info, and I won't make assumptions
giving you fifty things to look for. Here's a quick one
though -

does the destination have a route BACK to your
firewall/subnet?
