Link second hard drive?

I've run out of space on my hard drive, but I have a second one (internal)
with plenty of space. I've already moved as many data files as I can to the
second one. I could unistall some applications, and reinstall them on the
second drive, but do I have to do something to make sure they work properly
across the line from one drive to another?

Basically you've thought it through and answered your own question already.
If you uninstall and re-install correctly and the program allows you to
specify the disk to be used then all should be good to go. The re-install
takes care of setting up correct registry entries and paths etc.
 
But I've heard that many (most?) applications need certain registry files
that don't necessarily come with that application (but are resident on the C
drive), and when you install an app on a drive other than "C," it can't find
those necessary files and won't work properly.

Is there anything to this?

For the most part the programs work fine when installed on a drive other than C. On my personal computer, I keep the operating system on C, install applications onto D, and put data on E. It works fine for me. Some programs indeed keep bits and pieces of themselves on C even when installed elsewhere. However, the program installation routine takes care of letting the system know where to find all its parts. Uninstalling the original program and then reinstalling it on another drive should take care of it. You may have to select a custom installation, however.
