I have successfully set up second (sub) account for my wife with her own
email address. However, messages to that address are still appearing
alongside the messages for my original main email address, within the overall
list of messages received. How do I set up an Outlook screen only for that
email address - separated from the original screen for the original "master"
account?

that depends on several factors. The most important one being, are your
email addresses truely separate, or is your wife's address an ALIAS of the
main account? What I mean is this:
Are your email addresses in the format of (e-mail address removed) and (e-mail address removed)
or are they in the format of (e-mail address removed) and
(e-mail address removed)?

If the former, then you have two options - either set up a second Outlook
profile for your wife, or, (and this is the best option), if you are using
either Windows 2000 or Windows XP set up a new User log-in for your wife.
if the latter, you can only use a Rule to separate the mails coming from the
second alias to a separate email folder.
