How do I correct an error message that says "permission denied"?

I have made changes in the local network and when I try to send it to the
remote website and publish, and get different errors - the first part of the
message is based on the files like "550 Family Focus Eyecare.htm:permission
denied" or "550 Darlene.jpg:permission denied". So basically I am unable to
publish anything to the website.

You need to contact your web host, as 500 errors come from the remote server.
