Hosts file reloading

When adding entries to the hosts file what command do you run without
restarting the PC.

There are Host file editors available for free.
Example, http://www.funkytoad.com/content/view/13/31/
You can edit and save the file and that's it, you do not have to Run
anything.

Once you've changed it, you're done. No need to restart or anything else.
It's referenced on the fly.
