In case of a stand-alone server and inheritance is not disabled, are the
local security settings(found in secpol.msc) updated by group policies
security settings(found in gpedit.msc)?

If yes, does this mean that any change made to local security settings will
get overriden the next time group policy is applied?

They are not overwritten physically. If a setting in the local policy is
also set in a higher level policy the higher level is effective
