Yes you can convert it, look at the "Convert" command.

The process is supposed to be quite safe. However, I personally suggest
taking a backup of any vital data before performing this or any other
similar major operation. Its far better to have a un-needed backup than it
is to have no files and regrets.

You do not have to reformat to convert from FAT32 to NTFS, but it is a one
way trip. Here is a KB article that describes the process:
