Error trying to open programs

2 of my programs will not open, a window comes up with "16 Bit Windows
Subsystem" in the heading.
The error message is " C;\WINDOWS\SYSTEM32\AUTOEXEC.NT. The system file is
not suitable for running MS-DOS and Microsoft Windows applications. Choose
'Close' to terminate the application.

Can anyone help?
