error running program

I have been running an MSDOS program on my computer for a few years, recently
I can't open the program . The program is medical billing program called
medisoft
I get a message as follows.
C:\windows|system32\autoexec.nt. the system file is not suitable for running
MS-Dos and microdoft windows applications.

All other programs work fine except for the one in question.
Any help would be greatly appreciated.
