Files which are in use (running programs, system files, etc) cannot be defragged. You might want to try the command line version. Open a command prompt window and enter DEFRAG /? for the list of command line options.

Have already tried the command way. Didn't work. Any other suggestions?? Thank

If you haven't defragged in a while, it can take quite a while for the process to complete. Can you be more specific than "didn't work"?
