My boss has dozens of desktop icons, mostly going to Word or PDF
files. Is there a way I can check which ones are still valid, without
manually opening each one?

I think the Desktop Cleanup Wizard only checks for icons pointing to
apps.

Really only your boss can determine what data files are "valid".

Create a folder on the desktop and move all data files within that desktop folder.

Download Ccleaner

http://www.ccleaner.com/

Please note there is a bug in this version ZA detects Ccleaner as
Virus, but it is a false positve

When you run Ccleaner click on Cleaner and Under System check Desktop
Shortcuts, Click On Analize and finally Run Cleaner

these way the program checks which ones are still valid, if is valid
keep it in your desktop, if not is deleted
