Never tried putting DOS without third party tools but it should be possible.
1. Plug USB disk.
2. Boot device from DOS floppy.
3. use format X: /S on USB disk. (X: replace with volume of USB)

Actually I'm not thinking anything. I can boot XPe and pass all FBA phases on USB disk. So this means that today's BIOS-es are
capable of booting from USB
Also, depending on your boot speed requirement you will need to make sure that USB 2.0 "High-Speed" Boot option is also supported by
BIOS. AFAIK, Phoenix and latest AWARD BIOS-es have it.
Hi - I've tried to create a bootable disk on key (2GB) and load it with XPE
or XP. While I can boot the DOK with Linux and FreeDOS, I have failed to get
XP to install. The explanation about formating the USB drive from a DOS
console using the /s option worked for me today.
