Why it isn't possible to set "Start Date" in Task custom
form as required filed?

Because you can't modify the first page of a task form. You would have
to replicate the first page on a custom page, adding external
date/time pickers since the internal ones won't be available. Then you
could set a validation property on the start date requiring it to be
filled in.
