Shortcut Key

As you know you can define a shortcut key to run the programs by pushing
some keys on your keyboard ,but I couldn't do this even a once ,I mean
doesn't work ! what would be the cause ?

Because the shortcut isn't on the desktop or start menu. You chose a key that an open application is using. Apps take precedance over shortcut keys. You removed the shortcut arrow from shortcuts in one of the two ways.
