Security tab on Windows XP Folder

How do I turn on the security tab on a folder's properties in XP Pro so I
can prevent people sharing a computer from seeing a file? I have done it
before, but can't remember how to access the function.

Change FAT32 partition to NTFS.

As David said, you must be running NTFS as your file system. Additionally, in Windows Explorer, go to Tools, Folder Options, View. Scroll down and uncheck "Use simple file sharing".
