According to Adobe Premiere, when I try to download a DV
movie, the maximum file size have been reached after 18
minutes. My hard disk it runs on is a FAT32 system. How
can I change it into a NTFS system - where do I find the
setup program, do I have to clean the disk first, or will
it keep all my information on the disk when it converts?
What are the risks of changing the disk to the new format?

From the command line (Start/Run/cmd.exe):

convert drivelettergoes here /fs:ntfs

You don't need to clean up the drive as long as you have enough space on it
for the conversion - in case you don't, the utility will let you know.

Converting to NTFS is straightforward.
However, I do not see how this has anything to do
with the Adobe Premiere issue.
Files in FAT32 have a larger maximum size that
in NTFS as these two are implemented in XP.
In any case, the maximum file size for either is
probably larger than your hard disk space.
