NTFS Conversion?--Let Me Put It Another Way

If I have a DVD image file on a NTFS partition (drive E) and I start
Nero from Drive D which is the active partition containing XP (but
FAT32) will I be able to burn it? Thanks.

Yes.

The file system used on the boot drive has no effect on the ability of
Windows XP to work with either FAT32 or NTFS partitions. They can be
mixed on the same physical drive without difficulty.
