Trying to choose between the Scan Elite 5400 II and Nikon V ED for
predominantly 35 mm colour neg scanning. Any actual user comments?

I have the 5400 which is great. Given the LED source on the Mk II and
faster scan rates (claimed), that alone makes it better. Lower price too.

In the end, either will do the job very well.

As a 5400 first gen user, I'd say the second gen sounds to be moving
towards the Nikon, in terms of light source and speed. Your choice
might come down to the difference in the film transport.
