how to enable PowerPoint pen on my slide ?

Jefi,
During show press Ctrl+P to show the pen, Ctrl+A to shows the arrow again,
Ctrl+M to show or hide the ink you applied.
Be aware that you will get a very small dot, easy to miss.
An alternative would be to use the pop-up toolbar at the bottom left of your
screen, you can click on the second button and choose between pen, marker
and such. If the toolbar does not pop up, go to Tools - options - on the
view tab check show pop-up toolbar.

Jeff,
Could you tell us what version of PowerPoint you have?
What happens if you choose the pop-up toolbar and choose the pen, drag on
the slide? Maybe you are using white ink?
Also check if the Microsoft handwriting component is correctly installed on
your computer.
Run setup from office cd, select Add or Remove Features, choose advanced
customization of applications. In the Office Shared Features check if
Microsoft Handwriting Component is selected to run from your computer. This
is required to be installed to let you use the pen in slide show view.
