I have a high volume printing machine that is capable of printing different
spot colors with different passes through the machine. I would like to be
able to to print a master from Word that displays only one of the colors in
the document. For example print just the Blue first and then on a second
master print the black.

MS Word is not the right tool to produce color separations; MS Publisher
apparently has a Color Separation feature.

What input does your printing machine accept? Postscript? Then it should
be able to generate color separations from that.

Or you can print to a Postscript (.PS) file, for which there are tools
to produce color separations.
