This has to be determined by your hardware.

Can you put RAM in your motherboard in that configuration? Have you checked
the manuals?
Is the 256MB stick compatable with your motherboard? Does it work alone?

what type of ram is it ... there is sdram and ddram ,
also there different speed like 100MHz , 133 and so
on ... your problem probably is the speed of it , if you
have an older computer its most likely can just handle
100MHz and you probably have 256 MB PC133 SDRAM or
DDRAM , would this be correct ??? If you know what type
your other Ram is then try to match with the same speed
and just get higher stick... I would bet you could use
256MB 100MHz... What type does your new one say 256MB
whatelse ???

On many PC's, depending on the OS and age of the PC, you
will have to install matching RAM chips - 64 + 128 = 192.
You might have to buy a 2nd 256 chip and pull out the old
64/128 and install 2 256 chips.

Mixtures can be a problem: see the motherboard specs for what is and is
not allowed - eg biggest module may need to be in specific slot. Also
getting those modules to make good contact is *not* that easy. The test
is that the initial memory count as you boot must pick it up, and be
seen in BIOS setup; without that it will not be reported to Windows
