Error message when loading new software

Every time i try to load a new piece of software e.g. game
etc. i get the follwoing fault message;-
16 bit windows subsystem
C:\windows\system32\autoexec.nt. The system file is not
suitable for running MS-Dos and Microsoft applications.
Choose clsoe to terminate the application.

i cannot load any games which is driving me mad, can
anyone help.
